RTiterFast® Adjuvant is a pH-neutral aqueous adjuvant with a bluish transparent appearance. Its primary component is a high-molecular-weight polymer, whose hydrophobic ends bind to protein antigens (including peptides or small molecules conjugated to carrier proteins) without altering their structure. Once injected into the animal, the adjuvant slowly releases the bound antigen, gradually stimulating the immune system. All auxiliary materials meet human-use pharmaceutical standards, ensuring that the adjuvant itself is non-toxic, non-immunogenic, and highly biocompatible. The adjuvant significantly reduces adverse reactions, such as inflammation, granulomas, or injection site necrosis, compared to Freund’s adjuvant. Furthermore, RTiterFast® contains no protein components, preventing antibody production against the adjuvant itself. By reducing antigen consumption and shortening the immunization cycle, it effectively lowers antibody production costs. RTiterFast® is primarily suitable for use in rabbits.
RTiterFast® Adjuvant has been tested in 1972 New Zealand White rabbits with various antigen types, including recombinant proteins, small molecules, peptides, inactivated whole viruses, and virus-like particles (VLPs). The adjuvant is ideal for developing polyclonal antibodies and Signal B monoclonal antibodies in rabbits. Over 1,000 high-quality antibodies have been successfully developed using RTiterFast®, with applications in WB, IHC, IP, IF, Sandwich ELISA, indirect ELISA, Neutralization assays, and more. The graph shows the serum titer distribution of 1972 New Zealand White rabbits immunized three times with RTiterFast®. Approximately 83% of the antigens used were recombinant proteins, 16% were peptides, and 1% were small molecules, inactivated whole viruses, or VLPs. Cutoff OD = 0.25.

Usage Recommendations
Immunization Protocol: Three immunizations are recommended (Day 0, Day 7, Day 14). Serum can be collected on Day 21 for titer evaluation. If titers are lower than expected, administer a booster on Day 21 or 22.
Antigen Application and Recommended Dosage:
1) For protein antigens, inactivated whole viruses, whole bacteria, or virus-like particles (VLPs): Use 180-220 μg per rabbit per dose for the first immunization, diluting the antigen to 300 μL and mixing with 100 μL of RTiterFast® before injection. Subsequent doses require 80-120 μg per rabbit per dose, prepared in the same way.
2) For small molecules or peptides conjugated to carrier proteins: Use 400-500 μg per rabbit per dose for the first immunization, diluting the antigen to 300 μL and mixing with 100 μL of RTiterFast® before injection. Subsequent doses require 250 μg per rabbit per dose, prepared in the same way.
Injection Methods: Inject 400 μL of the antigen-adjuvant mixture per rabbit. Common injection methods include subcutaneous multi-point injections, intraperitoneal injections, and intramuscular injections. Intramuscular injection into the calf is recommended for convenience.
Preparation Method: Dilute the antigen with sterile saline or PBS to the recommended volume based on the antigen usage and the planned number of animals. Mix with the required amount of adjuvant and inject immediately after thorough mixing.
Storage Conditions: Store at 4-8°C. Avoid freezing. Shelf life: Two years.
Precautions
1) Precipitation after mixing with antigen is normal. Mix thoroughly before loading into the syringe and inject immediately after loading.
2) The adjuvant is optimized for use in rabbits and may have slightly reduced efficacy in other species.
3) Handle with care when opening to avoid injury from the aluminum seal.
